Freezing is the first stage and sets the ice structure that later becomes the pore network. The formulation is cooled below its freezing point, often with a controlled ramp, and solutes concentrate as ice forms. Primary drying then lowers chamber pressure and supplies heat to sublime the ice. The product temperature must stay below its collapse or eutectic temperature to prevent structural loss. Secondary drying raises the temperature modestly to remove bound water and achieve a low residual moisture.
A freeze-dryer consists of a vacuum chamber, temperature-controlled shelves, a condenser, and a vacuum pump. Vials, ampoules, or bulk trays hold the product during the cycle. The condenser traps water vapor as ice at a temperature lower than the product. Cycle development balances shelf temperature, chamber pressure, and time. Scale-up can be difficult because heat and mass transfer change with equipment size, so process analytical tools and conservative validation are often used.
Freeze-dried materials are hygroscopic to varying degrees and can take up moisture after drying. Storage therefore often uses sealed glass vials, rubber stoppers, and crimp seals to limit contact with ambient humidity. A desiccant may be included for moisture-sensitive products, although it is not universal. Controlled room temperature is sufficient for many lyophilizates, while others require refrigeration or freezing. Moisture ingress remains a primary cause of cake collapse, chemical degradation, and loss of reconstitution performance.
Quality assessment of a lyophilized product includes cake appearance, residual moisture, reconstitution time, and container closure integrity. A uniform, porous cake suggests that freezing and drying stayed within the formulation's design space. Cracks, shrinkage, meltback, or a glassy film can indicate thermal abuse or a formulation problem. Analysts also test for subvisible particles and sterility when the product requires those specifications. Visual inspection alone cannot confirm biological activity or chemical stability, so it is combined with analytical methods.

A steady state, on the other hand, is not necessarily an equilibrium state in the chemical sense. For example, in a radioactive decay chain the concentrations of intermediate isotopes are constant because the rate of production is equal to the rate of decay. It is not a chemical equilibrium because the decay process occurs in one direction only. Thermodynamic equilibrium is characterized by the free energy for the whole (closed) system being a minimum. For systems at constant volume the Helmholtz free energy is minimum and for systems at constant pressure the Gibbs free energy is minimum. Thus a metastable state is one for which the free energy change between reactants and products is not minimal even though the composition does not change in time. The existence of this minimum is due to the free energy of mixing of reactants and products being always negative. For ideal solutions the enthalpy of mixing is zero, so the minimum exists because the entropy of mixing is always positive. The slope of the reaction free energy, δGr with respect to the reaction coordinate, ξ, is zero when the free energy is at its minimum value.

In the late 1930s, questions emerged from League of Nations' Opium Advisory Committee concerning the focus on drug prohibition over public health measures such as mental health treatment, drug dispensaries and education. Anslinger, backed by his Canadian counterpart and policy ally, Charles Henry Ludovic Sharman, successfully argued against this view, and kept the focus on increasing global prohibition and supply control measures. While narcotics were under the jurisdiction of the FBN, the Federal Food, Drug, and Cosmetic Act of 1938 required the FDA to ensure that non-narcotic drugs were labeled for safe use. The act determined that certain drugs, including amphetamines, commercialized in the later 1930s, and barbiturates, were unsafe to use without medical supervision and could only be obtained by doctor's prescription. This marked the beginning of the federal distinction between over-the-counter and prescription drugs (clarified in the Durham–Humphrey Amendment of 1951).

Independence also did not result in stable political regimes, save in a few countries. First, the new nations did not have well-defined identities; rather, the process of creating identities was only beginning. This process would be carried out through newspapers and the creation of national symbols, including new names for the countries ("Mexico", "Colombia", "Ecuador", "Bolivia", "Argentina"), that broke with the past.
