Quality control for lyophilized products includes appearance, cake structure, reconstitution time, pH, residual moisture, and potency. Residual moisture is a key attribute because excess water can reduce stability, while excessively low moisture may cause structural changes or aggregation in some systems. Stability studies compare real-time and accelerated conditions to estimate shelf life. Analytical methods must be validated for the specific matrix, container, and moisture range. Sterility and container integrity are also monitored for sterile products.
Handling practices aim to prevent moisture ingress and mechanical damage. Vials should remain stoppered and crimped until use, and reconstitution should follow the labeled diluent and volume. Shipping may involve temperature-controlled containers and desiccants, but direct contact between desiccant and product is avoided. Regulatory guidance expects documented storage conditions, excursion assessments, and stability commitments. Open questions remain about how best to predict long-term stability from short accelerated studies for every formulation class.
After lyophilization, the product is usually a porous cake or powder with a large internal surface area. This structure can absorb moisture quickly if exposed to humid air, so vials are sealed under vacuum or an inert gas. Moisture uptake may lower the glass transition temperature of the dried matrix and accelerate chemical or physical degradation. Storage conditions therefore depend on the formulation, container, and intended shelf life. Some products remain stable at room temperature, while others require refrigeration or freezing.
After drying, a lyophilized product is usually sealed under vacuum or an inert gas to limit moisture and oxygen exposure. The container closure system matters because stoppers and seals can allow moisture ingress over time. Storage conditions are selected from stability studies that track potency, cake appearance, and reconstitution behavior. Many freeze-dried materials are kept at controlled room temperature, while some require refrigeration or protection from light.
Quality control for freeze-dried forms includes visual inspection, water content measurement, and reconstitution time. A satisfactory cake is typically uniform, porous, and intact, although minor shrinkage or cracking may be acceptable if specifications allow. Karl Fischer titration, thermal gravimetric analysis, and near-infrared spectroscopy are used to measure water content. Reconstitution is assessed by adding a specified diluent and recording the time and ease of dissolution. Microbiological and particulate tests are added when the product is sterile or intended for injection.

|---|---|---|
| Typical appearance | White to off-white porous cake or powder | Color and structure vary with formulation. |
| Typical reconstitution time | Seconds to several minutes | Diluent, agitation, and temperature affect rate. |
| Typical storage temperature | 2–8 °C, 15–25 °C, or ≤−20 °C | Product-specific; protect from moisture and light. |
| Typical container closure | Glass vial with rubber stopper and crimp seal | Closure must limit moisture ingress. |
| Typical stability indicator | Residual moisture, potency, and reconstitution time | Monitored throughout shelf life. |
Storage conditions depend on the formulation and the intended shelf life. Many pharmaceutical and biological freeze-dried products are kept at 2–8 °C, while some stable foods and reagents tolerate room temperature. Others require −20 °C or colder to slow chemical degradation or aggregation. Protection from light and oxygen is common because oxidation can continue in the dry state. Stability studies usually monitor potency, appearance, moisture, and reconstitution time over months or years. Predictions from accelerated studies are useful but may not fully capture real-time changes.
Quality control for freeze-dried lots combines visual inspection with instrumental tests. Cake appearance, color, and shrinkage are recorded against a reference, while residual moisture is measured by Karl Fischer titration or loss on drying. Thermal analysis can reveal phase transitions and crystallization events, and X-ray diffraction distinguishes amorphous from crystalline solids. Microbiological tests and container closure integrity checks are also routine for sterile products. Analytical methods must be validated for the matrix, because excipients and low moisture can affect accuracy. Open questions include how best to predict long-term stability from short-term data.
Lyophilized solids are often hygroscopic, so handling occurs in controlled low-humidity areas or glove boxes when the material is exposed. Vials remain sealed with elastomeric stoppers and aluminum crimps until use, because airborne moisture can raise residual water and shorten shelf life. The porous cake is fragile and may crack, shrink, or powder during transport. Personnel typically avoid repeated warming and cooling of sealed units, which can draw moisture through closures. These practices aim to preserve the low water content achieved during drying.
Handling and storage practices aim to keep the cake intact and dry. Vials are typically stored upright at controlled temperatures, often between 2 °C and 8 °C or at -20 °C for longer-term use. Reconstitution involves adding a suitable diluent and gently mixing until the solid dissolves. Shaking or rapid injection of diluent can create foam or damage sensitive molecules. Once reconstituted, the product may require refrigeration and use within a defined period.
Residual moisture is a key quality attribute for lyophilized products. Water that remains after secondary drying can affect chemical stability, cake structure, and shelf life. Karl Fischer titration is a common method for measuring water content in the dried solid. The target range varies by product, but many biologics are dried to between 0.5% and 3% water by weight. Acceptable limits are set during development and confirmed by stability studies.
Stability studies examine how temperature, humidity, and time influence a lyophilized product. Accelerated conditions provide early information about degradation pathways, while long-term studies support shelf-life claims. The glass transition temperature of the dried formulation can indicate its physical stability, and storage above this temperature may increase molecular mobility and lead to collapse or aggregation. Container closure integrity also matters because moisture or oxygen ingress can degrade the product, so vial stoppers and seals are part of the quality system.
Regulatory expectations for lyophilized products focus on consistent manufacture and documented stability. Batches are often monitored for moisture, appearance, potency, and sterility where applicable. Process parameters such as shelf temperature, chamber pressure, and drying time are recorded and controlled within validated ranges. Open questions remain about how best to predict long-term stability from short accelerated studies, especially for complex biologics. Variations in freezing rate and ice crystal size can produce differences that are not always visible but may affect performance.
After drying, a lyophilized product is usually sealed under vacuum or an inert gas to limit moisture uptake. Residual water content is measured because small changes can alter chemical stability and cake appearance. Storage temperature depends on the material; many biological products are kept at 2–8 °C, while some require −20 °C or colder. Exposure to ambient humidity during handling can cause the porous solid to absorb water and collapse. Container closures and stoppers are therefore selected for low moisture transmission and compatibility.
Quality control also examines cake structure, color, and reconstitution behavior. A collapsed or shrunken cake can indicate a thermal excursion during drying. Analytical methods such as X-ray diffraction, differential scanning calorimetry, and near-infrared spectroscopy can detect crystallinity or moisture distribution. Regulatory expectations focus on validated assays and lot-to-lot consistency. Questions remain about how well accelerated stability tests predict long-term behavior for every formulation. Visual inspection remains common but is subjective without trained reviewers and reference images.
After lyophilization, a product's quality depends on residual moisture, cake appearance, and reconstitution time. Residual moisture is often measured by Karl Fischer titration or thermogravimetric analysis. A low moisture content can slow chemical degradation, but overly dry cakes may be brittle or slow to dissolve. Stability studies track these attributes over months under defined temperature and humidity conditions. Batch records link these measurements to specific process runs and help identify trends before a product fails specification.
Storage conditions for dried products usually aim to exclude moisture and oxygen. Vials are sealed under vacuum or with an inert gas, and stoppers must maintain a barrier during transport. Temperature recommendations vary; some materials remain stable at room temperature, while others need refrigeration or frozen storage. Humidity control is critical because dried cakes can absorb water rapidly once a container is opened. Desiccant packs and moisture-barrier bags add further protection during shipping.
In the United States, the FDA relaxed rules on prescription drug marketing in 1997, allowing advertisements targeted directly to consumers. Lilly-ICOS hired the Grey Worldwide Agency in New York, part of the Grey Global Group, to run the Cialis advertising campaign. Marketing for Cialis has taken advantage of its greater duration compared to its competitors in advertisements for the drug; Stuart Elliot of The New York Times opined: "The continuous presence of females in Cialis ads is a subtle signal that the drug makes it easier for them to set the pace with males, in contrast to the primarily male-driven imagery for Levitra and Viagra." Iconic themes in Cialis ads include couples side by side in matching bathtubs and the slogan "When the moment is right, will you be ready?" Cialis ads were unique among the ED drugs in mentioning specifics of the drug. As a result, Cialis ads were also the first to describe the side effects in an advertisement, as the FDA requires advertisements with specifics to mention side effects. One of the first Cialis ads aired at the 2004 Super Bowl. Just weeks before the Super Bowl, the FDA required more possible side effects to be listed in the advertisement, including priapism. Although many parents objected to the Cialis ad being aired during the Super Bowl, Janet Jackson's halftime "wardrobe malfunction" overshadowed Cialis. In January 2006, the Cialis ads were tweaked, adding a doctor on screen to describe side effects and only running ads where more than 90 percent of the audience are adults, effectively ending Super Bowl ads.
However, the original cold noodle dish was not soft enough, and was a little hard to chew. After the use of softer noodles, grilled cold noodles was gradually accepted by people all around China. However, grilled cold noodles are still known for their chewiness. The chewy texture comes from gluten.

== History == Diclofenac was first synthesized by Alfred Sallmann and Rudolf Pfister in 1973. The name "diclofenac" derives from its chemical name: 2-(2,6-dichloranilino) phenylacetic acid. It was patented in Germany in 1978 by Ciba-Geigy (now Novartis). It came into medical use in the United States in 1988. GlaxoSmithKline purchased the rights in 2015. It is available as a generic medication.
== Clinical trial == The National Institutes of Health announced on 27 February 2015 the commencement of a randomized controlled trial of ZMapp to be conducted in Liberia and the United States. From March 2015 through November 2015, 72 individuals infected with the Ebola virus were enrolled in the trial; investigators stopped enrolling new subjects in January 2016, having failed to reach its enrollment goal of 200 due to the waning of the Ebola outbreak. As a result, although a 40% lower risk of death was calculated for those who received ZMapp, the difference was not statistically significant and ultimately it could not be determined whether the use of ZMapp was superior to the optimized standard of care alone. However, ZMapp was found to be safe and well tolerated.
== Toxicity == One review on graphene toxicity published in 2016 by Lalwani et al. summarizes the in vitro, in vivo, antimicrobial and environmental effects and highlights the various mechanisms of graphene toxicity. Another review published in 2016 by Ou et al. focused on graphene-family nanomaterials (GFNs) and revealed several typical mechanisms such as physical destruction, oxidative stress, DNA damage, inflammatory response, apoptosis, autophagy, and necrosis. A 2020 study showed that the toxicity of graphene is dependent on several factors such as shape, size, purity, post-production processing steps, oxidative state, functional groups, dispersion state, synthesis methods, route and dose of administration, and exposure times. In 2014, research at Stony Brook University showed that graphene nanoribbons, graphene nanoplatelets, and graphene nano–onions are non-toxic at concentrations up to 50 μg/ml. These nanoparticles do not alter the differentiation of human bone marrow stem cells towards osteoblasts (bone) or adipocytes (fat), suggesting that at low doses, graphene nanoparticles are safe for biomedical applications. In 2013, research at Brown University found that 10 μm few-layered graphene flakes can pierce cell membranes in solution. They were observed to enter initially via sharp and jagged points, allowing graphene to be internalized in the cell. The physiological effects of this remain unknown, and this remains a relatively unexplored field.
==== Motile sperm organelle morphology examination ==== A motile sperm organelle morphology examination (MSOME) is a particular morphologic investigation wherein an inverted light microscope equipped with high-power optics and enhanced by digital imaging is used to achieve a magnification above x6000, which is much higher than the magnification used habitually by embryologists in spermatozoa selection for intracytoplasmic sperm injection (x200 to x400). A potential finding on MSOME is the presence of sperm vacuoles, which are associated with sperm chromatin immaturity, particularly in the case of large vacuoles.

When a patient develops a simultaneous direct and indirect hernia on the same side, it is called a pantaloon hernia or saddlebag hernia because it resembles a pair of pants with the epigastric vessels in the crotch, and the defects can be repaired separately or together. Another term for pantaloon hernia is Romberg's hernia. Since the abdominal walls weaken with age, direct hernias tend to occur in the middle-aged and elderly. This is in contrast to indirect hernias, which can occur at any age, including the young, since their etiology includes a congenital component where the inguinal canal is left more patent (compared to individuals less susceptible to indirect hernias). Additional risk factors include chronic constipation, being overweight or obese, chronic cough, family history and prior episodes of direct inguinal hernias.
Vaginal discharge may indicate a vaginal infection by color and odor, or the resulting symptoms of discharge, such as irritation or burning. Abnormal vaginal discharge may be caused by STIs, diabetes, douches, fragranced soaps, bubble baths, birth control pills, yeast infection (commonly as a result of antibiotic use) or another form of vaginitis. While vaginitis is an inflammation of the vagina, and is attributed to infection, hormonal issues, or irritants, vaginismus is an involuntary tightening of the vagina muscles during vaginal penetration that is caused by a conditioned reflex or disease. Vaginal discharge due to yeast infection is usually thick, creamy in color and odorless, while discharge due to bacterial vaginosis is gray-white in color, and discharge due to trichomoniasis is usually a gray color, thin in consistency, and has a fishy odor. Discharge in 25% of the trichomoniasis cases is yellow-green. HIV/AIDS, human papillomavirus (HPV), genital herpes and trichomoniasis are some STIs that may affect the vagina, and health sources recommend safe sex (or barrier method) practices to prevent the transmission of these and other STIs. Safe sex commonly involves the use of condoms, and sometimes female condoms (which give women more control). Both types can help avert pregnancy by preventing semen from coming in contact with the vagina.
On the other hand, a fast reactor needs no moderator to slow down the neutrons at all, taking advantage of the fast neutrons producing a greater number of neutrons per fission than slow neutrons. For this reason ordinary liquid water, being a moderator and neutron absorber, is an undesirable primary coolant for fast reactors. Because large amounts of water in the core are required to cool the reactor, the yield of neutrons and therefore breeding of 239Pu are strongly affected. Theoretical work has been done on reduced moderation water reactors, which may have a sufficiently fast spectrum to provide a breeding ratio slightly over 1. This would likely result in an unacceptable power derating and high costs in a liquid-water-cooled reactor, but the supercritical water coolant of the supercritical water reactor (SCWR) has sufficient heat capacity to allow adequate cooling with less water, making a fast-spectrum water-cooled reactor a practical possibility. The type of coolants, temperatures, and fast neutron spectrum puts the fuel cladding material (normally austenitic stainless or ferritic-martensitic steels) under extreme conditions. The understanding of the radiation damage, coolant interactions, stresses, and temperatures are necessary for the safe operation of any reactor core. All materials used to date in sodium-cooled fast reactors have known limits. Oxide dispersion-strengthened alloy steel is viewed as the long-term radiation resistant fuel-cladding material that can overcome the shortcomings of today's material choices.
== Mechanism of action == Dalbavancin is a lipoglycopeptide belonging in the same glycopeptide class as vancomycin. Similar to other glycopeptides, dalbavancin exerts its bactericidal effect by disrupting cell wall biosynthesis. It binds to the D-alanyl-D-alanyl residue on growing peptidoglycan chains and prevents transpeptidation from occurring, preventing peptidoglycan elongation and cell wall formation. Dalbavancin also dimerizes and anchors itself in the lipophilic bacterial membrane, thereby increasing its stability in the target environment and its affinity for peptidoglycan. Antimicrobial activity correlates with the ratio of area under the concentration-time curve to minimum inhibitory concentration for Staphylococcus aureus.
In Scotland, the pharmacist can write prescriptions for Scottish registered patients of their regular medications, for the majority of drugs, except for controlled drugs, when the patient is unable to see their doctor, as could happen if they are away from home or the doctor is unavailable. In the United States, pharmaceutical care or clinical pharmacy has had an evolving influence on the practice of pharmacy. Moreover, the Doctor of Pharmacy (Pharm. D.) degree is now required before entering practice and some pharmacists now complete one or two years of residency or fellowship training following graduation. In addition, consultant pharmacists, who traditionally operated primarily in nursing homes, are now expanding into direct consultation with patients, under the banner of "senior care pharmacy". In addition to patient care, pharmacies will be a focal point for medical adherence initiatives. There is enough evidence to show that integrated pharmacy based initiatives significantly impact adherence for chronic patients. For example, a study published in National Institute for Health (NIH) shows "pharmacy based interventions improved patients' medication adherence rates by 2.1 percent and increased physicians' initiation rates by 38 percent, compared to the control group".

Collapse occurs when the product temperature rises above its collapse or eutectic temperature during drying. The frozen matrix loses structure, producing a shrunken or melted appearance. This can slow reconstitution and may affect stability.
Karl Fischer titration is a common method, along with loss on drying and thermogravimetric analysis. Each method has different sensitivity and sample requirements. Results should be interpreted with the product's formulation and container in mind.
No. Lyophilization removes water but is not a sterilization step. Sterile products are typically filtered and filled aseptically before freezing, and container closure integrity is maintained afterward.
Sealed vials or containers should be kept at the temperature specified by stability data, often controlled room temperature or 2–8 °C. Moisture and oxygen barriers are important because both can degrade sensitive materials. Opened containers may need immediate use or protection from ambient humidity.
